Surface Sterilization Protocol for Nodal Cultures of Potato (Solanum tuberosum L.)

The process of surface sterilizing plant materials is a difficult step in constructing tissue culture technique. Nodal explants from potted plants Solanum tuberosum L. grown the screenhouse were given various sterilization procedures before being introduced into sterile Murashige and Skoog (MS) culture. A total 16 treatments performed with varied periods immersion two sterilant successions: 70% (v/v) ethanol (20 s, 1 min, 3 5 min) 10% (w/v) calcium hypochlorite (Ca(ClO)2) granules (5, 10, 15, 20 at varying times, followed by three distilled water rinses. After 4 weeks culture, 70 % s)+10 Ca(ClO)2 (15 (3 min)+10 yielded 100 clean seconds) Plus (ClO) 2 recommended as optimum protocol since it produced 90% growth compared to 60% + min). This preliminary study, more research needed develop an efficient, successful, repeatable methodology for utilized S. biafrae acquired open field.
